Abstract Recent proof-of-principle works demonstrated a possibility to detect single biological nano-objects by the means of modified SPR imaging technique. In current study, we characterize bio-analytical features of modified SPRi method. As we demonstrated, modified SPRi technique allows detection and visualization of single HIV-VLPs and inactivated influenza A (IAV) viral particles not only in buffers, but also in buffers containing different concentrations of serum (up to 50%). Furthermore, we investigated the dependence of signal counting rate on the density of antibodies onto the biosensor surface. We also showed the applicability of modified SPRi technique for the determination of VLP concentrations in buffers. Altogether, our findings revealed novel opportunities for SPRi technique in such research areas as viral biology and biology of extracellular vesicles (exosomes and microvesicles).
